Folate Metabolism Linked to Redox Balance in Arabidopsis.

چکیده

Folates are soluble B9 vitamins with essential functions in all kingdoms of life—both in organisms that produce these vitamins de novo (fungi, plants, and most microorganisms) and in those that do not (animals). As essential cofactors in one-carbon transfers, different folate species mediate the biosynthesis of multiple biomolecules, including nucleic acids, amino acids, and lipids. Folates also play key roles in the methylation of DNA, proteins, and lipids. With such fundamental roles, it is not surprising that deficiencies of this vitamin have severe consequences. Cellular folate levels must be tightly controlled to match supply with demand.

Methoxistasis: Integrating the Roles of Homocysteine and Folic Acid in Cardiovascular Pathobiology

Over the last four decades, abnormalities in the methionine-homocysteine cycle and associated folate metabolism have garnered great interest due to the reported link between hyperhomocysteinemia and human pathology, especially atherothrombotic cardiovascular disease. However, clinical trials of B-vitamin supplementation including high doses of folic acid have not demonstrated any benefit in pre...

Amino acid homeostasis modulates salicylic acid-associated redox status and defense responses in Arabidopsis.

The tight association between nitrogen status and pathogenesis has been broadly documented in plant-pathogen interactions. However, the interface between primary metabolism and disease responses remains largely unclear. Here, we show that knockout of a single amino acid transporter, LYSINE HISTIDINE TRANSPORTER1 (LHT1), is sufficient for Arabidopsis thaliana plants to confer a broad spectrum of...

The folylpolyglutamate synthetase plastidial isoform is required for postembryonic root development in Arabidopsis.

A recessive Arabidopsis (Arabidopsis thaliana) mutant with short primary roots and root hairs was identified from a forward genetic screen. The disrupted gene in the mutant encoded the plastidial isoform of folylpolyglutamate synthetase (FPGS), previously designated as AtDFB, an enzyme that catalyzes the addition of glutamate residues to the folate molecule to form folylpolyglutamates. The shor...

Human mitochondrial MTHFD2 is a dual redox cofactor-specific methylenetetrahydrofolate dehydrogenase/methenyltetrahydrofolate cyclohydrolase

Background Folate-dependent one-carbon metabolism provides one-carbon units for several biological processes. This pathway is highly compartmentalized in eukaryotes, with the mitochondrial pathway producing formate for use in cytoplasmic processes. The mitochondrial enzyme MTHFD2 has been reported to use NAD+ as a cofactor while the isozyme MTHFD2L utilizes NAD+ or NADP+ at physiologically rele...

نقش هوموسیستیین درتندرستی و بیماری

High plasma homocysteine is identified as an important risk factor for cardiovascular disease. Epidemiological studies have shown that increased plasma homocysteine is related to a higher risk of coronary heart disease, stroke and peripheral vascular disease. Homocysteine may have an effect on atherosclerosis by damaging the inner lining of arteries and promoting blood clots .Folic acid and oth...
